概述 Overview

本品为马钱科植物密蒙花Buddleja officinalis Maxim.的干燥花蕾和花序。春季花未开放时采收,除去杂质,干燥。
Buddleja Flower is the dried flower buds and inflorescences of Buddleja officinalis Maxim. (Fam. Loganiaceae). The drug is collected in spring before the flowers open, removed from impurities, and dried.

性状 Description

本品多为花蕾密聚的花序小分枝,呈不规则圆锥状,长1.5~3cm。表面灰黄色或棕黄色,密被茸毛。花蕾呈短棒状,上端略大,长0.3~1cm,直径0.1~0.2cm;花萼钟状,先端4齿裂;花冠筒状,与萼等长或稍长,先端4裂,裂片卵形;雄蕊4,着生在花冠管中部。质柔软。气微香,味微苦、辛。
The drug is composed of small branches of inflorescences with densely clustered flower buds, irregularly conical, 1.5-3 cm long. Surface grayish-yellow or brownish-yellow, densely covered with pubescence. Flower buds short and rod-shaped, slightly larger at the upper end, 0.3-1 cm long, 0.1-0.2 cm in diameter; calyx campanulate, 4-toothed at the apex; corolla tubular, about as long as or slightly longer than the calyx, 4-lobed at the apex, lobes ovate; stamens 4, inserted in the middle part of the corolla tube. Texture soft. Odour, slightly fragrant; taste, slightly bitter and pungent.

鉴别 Identification

本品粉末棕色。非腺毛通常为4细胞,基部2细胞单列;上部2细胞并列,每细胞又分2叉,每分叉长50~500μm,壁甚厚,胞腔线形。花冠上表面有少数非腺毛,单细胞,长38~600μm,壁具多数刺状突起。花粉粒球形,直径13~20μm,表面光滑,有3个萌发孔。腺毛头部顶面观(1~)2细胞,2细胞者并列呈哑铃形或蝶形;柄极短。
The powder is brown. Non-glandular hairs usually consist of 4 cells, with 2 cells at the base arranged singly and 2 cells at the upper part arranged in pairs, each cell bifurcating into 2 branches, each branch 50-500 μm long, with thick walls and linear cell cavities. The upper surface of the corolla has a few non-glandular hairs, consisting of single cells, 38-600 μm long, with numerous prickly projections on the walls. Pollen grains spherical, 13-20 μm in diameter, with smooth surface and 3 germination pores. The head of glandular hairs is composed of (1-)2 cells, with 2 cells arranged in pairs in the shape of a dumbbell or butterfly; the stalk is very short.

含量测定 Content Determination

照高效液相色谱法(通则0512)测定。
Carry out the method for high performance liquid chromatography <0512>.

色谱条件与系统适用性试验 Chromatographic conditions and system suitability test

以十八烷基硅烷键合硅胶为填充剂;以甲醇-水-醋酸(45︰54.5︰0.5)为流动相;检测波长为326nm。理论板数按蒙花苷峰计算应不低于1000。
Use octadecylsilane bonded silica gel as the stationary phase; use methanol-water-acetic acid (45:54.5:0.5) as the mobile phase; detect at a wavelength of 326 nm. The theoretical number of plates calculated based on the peak of verbascoside should not be less than 1000.

对照品溶液的制备 Preparation of reference solution

取蒙花苷对照品适量,精密称定,加甲醇制成每1ml含蒙花苷0.1mg的溶液,即得。
Take an appropriate amount of verbascoside reference substance, accurately weigh, dissolve in methanol to obtain a solution containing 0.1 mg of verbascoside per ml.

供试品溶液的制备 Preparation of test solution

取本品粉末约0.5g,精密称定,置索氏提取器中,加石油醚(60~90 ℃)100ml,加热回流2小时,弃去石油醚,药渣挥干,再加甲醇100ml继续加热回流4小时,提取液置蒸发皿中,浓缩至适量,转移至50ml量瓶中,残渣及容器用少量甲醇洗涤,洗液并入同一量瓶中,加甲醇至刻度,摇匀,滤过,取续滤液,即得。
Take about 0.5 g of the powder, accurately weigh, place it in a Soxhlet extractor, add 100 ml of petroleum ether (60-90°C), heat under reflux for 2 hours, discard the petroleum ether, dry the residue, then add 100 ml of methanol and continue to heat under reflux for 4 hours. Transfer the extract to an evaporating dish, concentrate to an appropriate volume, transfer to a 50 ml volumetric flask, wash the residue and the container with a small amount of methanol, combine the washings in the same volumetric flask, add methanol to the mark, shake well, filter, and take the filtrate to obtain the test solution.

测定法 Determination method

分别精密吸取对照品溶液10μl与供试品溶液5~10μl,注入液相色谱仪,测定,即得。
Accurately aspirate 10 μl of the reference solution and 5-10 μl of the test solution, inject into the liquid chromatograph, and determine.

本品含蒙花苷(C28H32O14)不得少于0.50%。
The content of verbascoside (C28H32O14) in the drug should not be less than 0.50%.

性状 Description

本品多为花蕾密聚的花序小分枝,呈不规则圆锥状,长1.5~3cm。表面灰黄色或棕黄色,密被茸毛。花蕾呈短棒状,上端略大,长0.3~1cm,直径0.1~0.2cm;花萼钟状,先端4齿裂;花冠筒状,与萼等长或稍长,先端4裂,裂片卵形;雄蕊4,着生在花冠管中部。质柔软。气微香,味微苦、辛。
Mainly small branches of inflorescence bearing dense flower buds, irregularly conical, 1.5-3 cm long. Externally greyish-yellow or brownish-yellow, densely pubescent. Flower buds clavate, with the upper side slightly larger. 0.3-1 cm long, 0.1-0.2 cm in diameter; calyx campanulate with 4 terminal teeth; corolla tubular, as long as calyx or longer, with 4 terminal lobes, lobes ovate; stamens 4; inserted on the middle of corolla tube. Texture soft. Odour; slightly aromatic; taste, slightly bitter and pungent.

Powder brown. Non-glandular hairs usually 4-celled, at the base 2-celled, uniseriate; at the apex 2-celled, biseriate, each cell 2-forked, forks 250-500 μm long, walls extremely thick, lumen linear. The upper epidermis of corolla with few non-glandular hairs; unicellular, 200-600 μm long, walls with numerous spiny emergences. Pollen grains globose, 13-20 μm in diameter; exine smooth, with 3 germinal pores.

The number of theoretical plates of the column is not less than 1000, calculated with reference to the peak of buddleoside.

取蒙花苷对照品适量,精密称定,加甲醇制成每1ml含蒙花苷0.1mg的溶液,即得。
Dissolve 10 mg, accurately weighed, in a 100 ml volumetric flask with methanol, dilute with methanol to volume, shake thoroughly to produce a solution of 0.1 mg per ml.

供试品溶液的制备 Test solution

取本品粉末约0.5g,精密称定,置索氏提取器中,加石油醚(60~90℃)100ml,加热回流2小时,弃去石油醚,药渣挥干,再加甲醇100ml继续加热回流4小时,提取液置蒸发皿中,浓缩至适量,转移至50ml量瓶中,残渣及容器用少量甲醇洗涤,洗液并入同一量瓶中,加甲醇至刻度,摇匀,滤过,取续滤液,即得。
Weigh accurately 0.5g of the powder in a Sohxlet's extractor, add 100 ml of petroleum ether (60-90℃); heat under reflux for 2 hours, discard petroleum ether and expel, add 100 ml of methanol and heat under reflux for 4 hours, concentrate the extracts to a quantity, transfer to a 50 ml volumetric flask, add methanol to volume.

测定法 Procedure

分别精密吸取对照品溶液10μl与供试品溶液5~10μl,注入液相色谱仪,测定,即得。
Inject separately 10 μl of the reference solution and 5-10 μl of the test solution, accurately measured, into the column, determine and calculate the content.

本品含蒙花苷(C28H32O14)不得少于0.50%。
It contains not less than 0.50 per cent of buddleoside (C28H32O14).

甘,微寒。归肝经。
Mild cold; sweet. Liver meridian.
