概述 Overview

本品为蚌壳蕨科植物金毛狗脊Cibotium barometz（L.）J.Sm.的干燥根茎。秋、冬二季采挖，除去泥沙，干燥；或去硬根、叶柄及金黄色绒毛，切厚片，干燥，为“生狗脊片”；蒸后晒至六、七成干，切厚片，干燥，为“熟狗脊片”。
Rhizoma Cibotii is the dried rhizome and root of Cibotium barometz (L.) J. Sm. (Fam. Dicksoniaceae). The drug is collected in autumn and winter, removed from sand and soil, and dried; or the hard root, petiole, and golden hairs are removed, cut into thick slices, and dried, known as "Raw Rhizoma Cibotii"; or steamed, dried until 60-70% dry, cut into thick slices, and dried, known as "Processed Rhizoma Cibotii".

性状 Description

本品呈不规则的长块状，长10～30 cm，直径2～10 cm。表面深棕色，残留金黄色绒毛；上面有数个红棕色的木质叶柄，下面残存黑色细根。质坚硬，不易折断。无臭，味淡、微涩。生狗脊片呈不规则长条形或圆形，长5～20 cm，直径2～10 cm，厚1.5～5 mm；切面浅棕色，较平滑，近边缘1～4 mm处有1条棕黄色隆起的木质部环纹或条纹，边缘不整齐，偶有金黄色绒毛残留；质脆，易折断，有粉性。熟狗脊片呈黑棕色，质坚硬。
The drug is irregularly long and block-shaped, 10-30 cm long, 2-10 cm in diameter. The surface is dark brown with residual golden hairs; there are several reddish-brown woody petioles on the upper surface, and black fine roots remain on the lower surface. The texture is hard and not easily broken. It has no odor, a mild taste, and a slightly astringent taste. Raw Rhizoma Cibotii is irregularly long or round, 5-20 cm long, 2-10 cm in diameter, and 1.5-5 mm thick; the cut surface is light brown, relatively smooth, with a brownish-yellow raised woody ring or stripe at 1-4 mm near the edge, the edge is irregular, and there may be residual golden hairs; the texture is brittle, easily broken, and has a powdery nature. Processed Rhizoma Cibotii is blackish-brown and has a hard texture.

鉴别 Identification

（1）本品横切面：表皮细胞1列，残存金黄色的非腺毛。其内有10余列棕黄色厚壁细胞，壁孔明显。木质部排列成环，由管胞组成，其内外均有韧皮部和内皮层。皮层和髓均由薄壁细胞组成，细胞充满淀粉粒，有的含黄棕色物。
（1）Transverse section: epidermal cells in one row, with residual golden non-glandular hairs. There are more than 10 rows of brownish-yellow thick-walled cells with obvious wall pores. The wood is arranged in rings, composed of vessel cells, with both phloem and endodermis inside and outside. The cortex and pith are composed of thin-walled cells, which are filled with starch grains, some of which contain yellowish-brown substances.

（2）取本品粉末2 g，加甲醇50 ml，超声处理30分钟，滤过，滤液蒸干，残渣加甲醇1 ml使溶解，作为供试品溶液。另取狗脊对照药材2 g，同法制成对照药材溶液。照薄层色谱法（通则0502）试验，吸取供试品溶液3～6 μl、对照药材溶液4 μl，分别点于同一硅胶G薄层板上，使成条状，以甲苯-三氯甲烷-乙酸乙酯-甲酸（3：5：6：1）为展开剂，展开，取出，晾干，喷以2%三氯化铁溶液-1%铁氰化钾溶液（1：1）（临用配制），放置至斑点显色清晰。供试品色谱中，在与对照药材色谱相应的位置上，显相同颜色的斑点。
（2）Take 2 g of the powder, add 50 ml of methanol, sonicate for 30 minutes, filter, evaporate the filtrate to dryness, dissolve the residue in 1ml of methanol as the test solution. Take 2 g of the reference drug, process it in the same way to obtain the reference drug solution. Carry out the method for thin layer chromatography <0502>, using silica gel G as the coating substance and a mixture of toluene-chloroform-ethyl acetate-formic acid (3: 5: 6: 1) as the mobile phase. Apply separately to the plate 3-6 μl of the test solution and 4 μl of the reference drug solution. After developing and removal of the plate, dry in air. Spray with a 2% solution of ferric chloride and a 1% solution of potassium ferricyanide (1: 1) (prepared before use). Leave until the spots are clearly visible. The fluorescent spot in the chromatogram obtained with the test solution corresponds in position and colour to the spot in the chromatogram obtained with the reference drug solution.

烫狗脊 ｜ Cibotii Rhizoma (scalded)

炮制 ｜ Processing

取生狗脊片，照炒法（通则0213) 用砂烫至鼓起，放凉后除去残存绒毛。
Scald the slices of Cibotii Rhizoma with sand as described under the method for scalding <0213> until inflated, and after cooling remove the remainder of hairs.

性状 Description

本品形如狗脊片，表面略鼓起。棕褐色。气微，味淡微涩。
The shape similar to the slices, surface slightly expanded, brown. Odour slight; taste, slightly astringent.

鉴别 ｜Identification

取本品粉末2 g，加甲醇50 ml，超声处理30 分钟，滤过，滤液蒸干，残渣加甲醇1 ml使溶解，作为供试品溶液。另取原儿茶醒对照品、原儿茶酸对照品，加甲醇制成每1 ml 各含0.5 mg 的混合溶液，作为对照品溶液。照薄层色谱法（通则0502) 试验，吸取供试品溶液3~6 μl 对照品溶液2 μl, 分别点于同一硅胶G薄层板上，使成条状，以三氯甲烷－乙酸乙酯甲醇－甲酸（12：2：1：0.8）为展开剂，展开，取出，晾干，喷以2%三氯化铁溶液-1%铁氰化钾溶液（1：1）（临用配制）。供试品色谱中，在与对照品色谱相应的位置上，显相同颜色的斑点。
To 2 g of the powder add 50 ml of methanol, ultrasonicate for 30 minutes, filter, evaporate the filtrate to dryness and dissolve the residue in 1 ml of methanol as the test solution. Dissolve protocatechuic aldehyde CRS and protocatechuic acid CRS in methanol to produce a mixture containing 0. 5 mg of each per ml as the reference solution. Carry out the method for thin layer chromatography <0502>, using silica gel G as the coating substance and a mixture of chloroform, ethyl acetate, methanol and formic acid (12: 2: 1: 0.8) as the mobile phase. Apply separately to the plate in strip 3-6 μl of the test solution and 2 μl of the reference solution. After developing and removal of the plate, dry in air, spray with a mixture of 2% solution of ferric chloride and 1% solution of potassium ferricyanide (1: 1, prepared before use). The spots in the chromatogram obtained with test solution correspond in position and colour to the spots in the chromatogram obtained with the reference solution.

以十八烷基硅烷键合硅胶为填充剂；以乙腈-1% 冰醋酸溶液（5：95）为流动相；检测波长为260 nm。理论板数按原儿茶酸峰计算应不低于3000。
Chromatographic conditions and system suitability test Octadecylsilane-bonded silica gel was used as filler; acetonitrile-1% glacial acetic acid solution (5: 95) was used as the mobile phase; and the detection wavelength was 260 nm; the theoretical plate number should be not less than 3000 according to the calculation of the original catechuic acid peak.

对照品溶液的制备 Preparation of reference solution

取原儿茶酸对照品适量，精密称定，加甲醇-1% 冰醋酸溶液（70：30）混合溶液制成每1 ml含50 μg的溶液，即得。
Weigh accurately a quantity of protocatechuic acid CRS, dissolve in a mixture of methanol and 1% solution of glacial acetic acid (70: 30) to produce a solution containing 50 μg per ml.

供试品溶液的制备 Preparation of test solution

取本品粉末（过三号筛）约1 g, 精密称定，置具塞锥形瓶中，精密加入甲醇-1% 冰醋酸溶液（70：30）混合溶液25 ml，称定重量，超声处（功率250 W，频率40 kHz)30分钟，放冷，再称定重量，用甲醇-1% 冰醋酸溶液（70：30）混合溶液补足减失的重量，摇匀，滤过，取续滤液，即得。
Weigh accurately 1 g of the powder to a stoppered conical flask, add accurately 25 ml of a mixture of methanol and 1% solution of glacial acetic acid (70: 30), weigh and ultrasonicate (power 250 W, frequency 40 kHz) for 30 minutes, cool and weigh again, replenish the loss of the solvent with the same mixture, mix well, filter and use the successive filtrate as the test solution.

测定法 Assay method

分别精密吸取对照品溶液与供试品溶液各10 μl, 注入液相色谱仪，测定，即得。
Inject accurately 10 μl of each of the reference solution and the test solution, respectively, into the column, and calculate the content.

本品按干燥品计算，含原儿茶酸(C7H6O4)不得少于0.020%。
This product contains not less than 0.020% protocatechuic acid (C7H6O4) calculated as dried product.

本品为蚌壳蕨科植物金毛狗脊Cibotium barometz （L.） J. Sm.的干燥根茎。秋、冬二季采挖，除去泥沙，干燥；或去硬根、叶柄及金黄色绒毛，切厚片，干燥，为“生狗脊片”；蒸后晒至六、七成干，切厚片，干燥，为“熟狗脊片”。
Cibot Rhizome is the dried rhizome of Cibotium barometz ( L.) J. Sm. ( Fam. Dicksoniaceae). The drug is collected in autumn or in winter, removed from soil and dried； or removed from hard root, frond base and hairs, thickly sliced, and dried, called “Shenggoujipian”； or after steaming the drug is partially dried in the sun, cut into thick slices and dried thoroughly, called “Shugoujipian”.

性状 Description

Irregularly long lump-shaped, 10-30 cm long, 2-10 cm in diameter. Externally dark brown, with remains of golden hairs； the upper part exhibiting several reddish-brown woody frond base and the lower part showing remains of black fibrous roots. Texture hard, uneasily broken. Odourless； taste, weak and slight astringent.
